    The effect of the color filter array layout choice on state-of-the-art demosaicing

    Interpolation from a Color Filter Array (CFA) is the most common method for obtaining full color image data. Its success relies on the smart combination of a CFA and a demosaicing algorithm. Demosaicing on the one hand has been extensively studied. Algorithmic development in the past 20 years ranges from simple linear interpolation to modern neural-network-based (NN) approaches that encode the prior knowledge of millions of training images to fill in missing data in an inconspicious way. CFA design, on the other hand, is less well studied, although still recognized to strongly impact demosaicing performance. This is because demosaicing algorithms are typically limited to one particular CFA pattern, impeding straightforward CFA comparison. This is starting to change with newer classes of demosaicing that may be considered generic or CFA-agnostic. In this study, by comparing performance of two state-of-the-art generic algorithms, we evaluate the potential of modern CFA-demosaicing. We test the hypothesis that, with the increasing power of NN-based demosaicing, the influence of optimal CFA design on system performance decreases. This hypothesis is supported with the experimental results. Such a finding would herald the possibility of relaxing CFA requirements, providing more freedom in the CFA design choice and producing high-quality cameras

    V-interESP - Utilização de Vídeos para Melhorar a Experiência de Aprendizagem dos Estudantes ESP: Ensino e Investigação Conjunta Internacional

    Addressing the growing need to re-examine current approaches and teaching methods within the scope of ESP, this paper starts by describing the preliminary states of V-interESP – an international joint research project involving students and faculty from Lithuania, Portugal and Serbia. This project has two primary goals: to understand the role digital technology, and particularly videos, can play in enhancing students’ learning experiences within ESP settings; and to promote the study participants’ creativity as well as intercultural awareness and communication. Thus, in this study, the authors present preliminary research findings, based on the students’ answers to an entry questionnaire. Data focusses on students’ profiles and their perceived potential of using video to learn ESP, in particular regarding student motivation and the development of communication skills, as well as creative abilities.    Tendo como principal objetivo rever abordagens e dinâmicas de ensino e aprendizagem no domínio do Inglês Para Fins Específicos, neste artigo descrevem-se as fases de planificação e implementação da primeira iteração do projeto V-interESP. Este projeto de investigação conjunta contou com a participação de alunos e docentes da Lituânia, Portugal e Sérvia, tendo com principais objetivos contribuir para a compreensão do papel da tecnologia digital, e mais especificamente dos vídeos, na promoção e enriquecimento das experiências dos alunos que frequentam aulas de Inglês para Fins Específicos; e promover a criatividade e a comunicação intercultural neste domínio. Desta forma, tendo por base um questionário inicial aplicado aos alunos, para além da descrição da implementação do projeto, são apresentados resultados preliminares quanto ao perfil dos participantes e as suas percepções, designadamente relativamente ao potencial da utilização de vídeos na aprendizagem de línguas e enquanto fator motivacional e de promoção de competências comunicativas e da criatividade

    Highlights Analysis System (HAnS) for low dynamic range to high dynamic range conversion of cinematic low dynamic range content

    We propose a novel and efficient algorithm for detection of specular reflections and light sources (highlights) in cinematic content. The detection of highlights is important for reconstructing them properly in the conversion of the low dynamic range (LDR) to high dynamic range (HDR) content. Highlights are often difficult to be distinguished from bright diffuse surfaces, due to their brightness being reduced in the conventional LDR content production. Moreover, the cinematic LDR content is subject to the artistic use of effects that change the apparent brightness of certain image regions (e.g. limiting depth of field, grading, complex multi-lighting setup, etc.). To ensure the robustness of highlights detection to these effects, the proposed algorithm goes beyond considering only absolute brightness and considers five different features. These features are: the size of the highlight relative to the size of the surrounding image structures, the relative contrast in the surrounding of the highlight, its absolute brightness expressed through the luminance (luma feature), through the saturation in the color space (maxRGB feature) and through the saturation in white (minRGB feature). We evaluate the algorithm on two different image data-sets. The first one is a publicly available LDR image data-set without cinematic content, which allows comparison to the broader State of the art. Additionally, for the evaluation on cinematic content, we create an image data-set consisted of manually annotated cinematic frames and real-world images. For the purpose of demonstrating the proposed highlights detection algorithm in a complete LDR-to-HDR conversion pipeline, we additionally propose a simple inverse-tone-mapping algorithm. The experimental analysis shows that the proposed approach outperforms conventional highlights detection algorithms on both image data-sets, achieves high quality reconstruction of the HDR content and is suited for use in LDR-to-HDR conversion

    Graphene-based materials have been extensively explored in recent years as valuable candidates as the key material for novel structures in the field, among many other applications, of sensing devices. Reduced Graphene Oxide (rGO) is a type of chemically derived graphene, with equivalent optical properties but easier to be synthetized. This work reports a study about the applicability of rGO as a support for gold nanoparticles (AuNPs).     Activation of Neurogenesis in Multipotent Stem Cells Cultured In Vitro and in the Spinal Cord Tissue After Severe Injury by Inhibition of Glycogen Synthase Kinase-3

    The inhibition of glycogen synthase kinase-3 (GSK-3) can induce neurogenesis, and the associated activation of Wnt/β-catenin signaling via GSK-3 inhibition may represent a means to promote motor function recovery following spinal cord injury (SCI) via increased astrocyte migration, reduced astrocyte apoptosis, and enhanced axonal growth. Herein, we assessed the effects of GSK-3 inhibition in vitro on the neurogenesis of ependymal stem/progenitor cells (epSPCs) resident in the mouse spinal cord and of human embryonic stem cell-derived neural progenitors (hESC-NPs) and human-induced pluripotent stem cell-derived neural progenitors (hiPSC-NPs) and in vivo on spinal cord tissue regeneration and motor activity after SCI. We report that the treatment of epSPCs and human pluripotent stem cell-derived neural progenitors (hPSC-NPs) with the GSK-3 inhibitor Ro3303544 activates β-catenin signaling and increases the expression of the bIII-tubulin neuronal marker; furthermore, the differentiation of Ro3303544-treated cells prompted an increase in the number of terminally differentiated neurons. Administration of a water-soluble, bioavailable form of this GSK-3 inhibitor (Ro3303544-Cl) in a severe SCI mouse model revealed the increased expression of bIII-tubulin in the injury epicenter. Treatment with Ro3303544-Cl increased survival of mature neuron types from the propriospinal tract (vGlut1, Parv) and raphe tract (5-HT), protein kinase C gamma-positive neurons, and GABAergic interneurons (GAD65/67) above the injury epicenter. Moreover, we observed higher numbers of newly born BrdU/DCX-positive neurons in Ro3303544-Cl-treated animal tissues, a reduced area delimited by astrocyte scar borders, and improved motor function. Based on this study, we believe that treating animals with epSPCs or hPSC-NPs in combination with Ro3303544-Cl deserves further investigation towards the development of a possible therapeutic strategy for SCI

    Expanding the importance of HMERF titinopathy : new mutations and clinical aspects

    ObjectiveHereditary myopathy with early respiratory failure (HMERF) is caused by titin A-band mutations in exon 344 and considered quite rare. Respiratory insufficiency is an early symptom. A collection of families and patients with muscle disease suggestive of HMERF was clinically and genetically studied.MethodsAltogether 12 new families with 19 affected patients and diverse nationalities were studied. Most of the patients were investigated using targeted next-generation sequencing; Sanger sequencing was applied in some of the patients and available family members. Histological data and muscle MRI findings were evaluated.ResultsThree families had several family members studied while the rest were single patients. Most patients had distal and proximal muscle weakness together with respiratory insufficiency. Five heterozygous TTN A-band mutations were identified of which two were novel. Also with the novel mutations the muscle pathology and imaging findings were compatible with the previous reports of HMERF.ConclusionsOur collection of 12 new families expands mutational spectrum with two new mutations identified. HMERF is not that rare and can be found worldwide, but maybe underdiagnosed. Diagnostic process seems to be complex as this study shows with mostly single patients without clear dominant family history.Peer reviewe

    Skeletal Muscle Biopsy Analysis in Reducing Body Myopathy and Other Fhl1-related Disorders

    FHL1 mutations have been associated with various disorders that include reducing body myopathy (RBM), Emery-Dreifuss-like muscular dystrophy, isolated hypertrophic cardiomyopathy, and some overlapping conditions. We report a detailed histochemical, immunohistochemical, electron microscopic, and immunoelectron microscopic analyses of muscle biopsies from 18 patients carrying mutations in FHL1: 14 RBM patients (Group 1), 3 Emery-Dreifuss muscular dystrophy patients (Group 2), and 1 patient with hypertrophic cardiomyopathy and muscular hypertrophy (Group 2). Group 1 muscle biopsies consistently showed RBs associated with cytoplasmic bodies. The RBs showed prominent FHL1 immunoreactivity whereas desmin, alpha B-crystallin, and myotilin immunoreactivity surrounded RBs. By electron microscopy, RBs were composed of electron-dense tubulofilamentous material that seemed to spread progressively between the myofibrils and around myonuclei. By immunoelectron microscopy, FHL1 protein was found exclusively inside RBs. Group 2 biopsies showed mild dystrophic abnormalities without RBs; only minor nonspecific myofibrillar abnormalities were observed under electron microscopy. Molecular analysis revealed missense mutations in the second FHL1 LIM domain in Group 1 patients and ins/del or missense mutations within the fourth FHL1 LIM domain in Group 2 patients. Our findings expand the morphologic features of RBM, clearly demonstrate the localization of FHL1 in RBs, and further illustrate major morphologic differences among different FHL1-related myopathies

    Antioxidant-inspired drug discovery: antitumor metabolite is formed in situ from a hydroxycinnamic acid derivative upon free radical scavenging

    Cancer cells generally possess higher levels of reactive oxygen species than normal cells, and this can serve as a possible therapeutic target. In this proof-of-concept study, an antioxidant-inspired drug discovery strategy was evaluated using a hydroxycinnamic acid derivative. The processing of oxidized mixtures of p-coumaric acid methyl ester (pcm) revealed a new antitumor lead, graviquinone. Graviquinone bypassed ABCB1-mediated resistance, induced DNA damage in lung carcinoma cells but exerted DNA protective activity in normal keratinocytes, and modulated DNA damage response in MCF-7 cells. The cytotoxic effect of pcm in MCF-7 cells was potentiated under H2O2-induced oxidative stress, and the formation of graviquinone was confirmed by Fenton's reaction on pcm. In silico density functional theory calculations suggested graviquinone as a kinetic product of pcm-scavenging (OH)-O-center dot radicals. Our results demonstrate the pharmacological value of an in situ-formed, oxidative stress-related metabolite of an antioxidant. This might be of particular importance for designing new strategies for antioxidant-based drug discovery

    Comparison of exercise, dobutamine-atropine and dipyridamole-atropine stress echocardiography in detecting coronary artery disease

    BACKGROUND: Dipyridamole and dobutamine stress echocardiography testing are most widely utilized, but their sensitivity remained suboptimal in comparison to routine exercise stress echocardiography. The aim of our study is to compare, head-to-head, exercise, dobutamine and dipyridamole stress echocardiography tests, performed with state-of-the-art protocols in a large scale prospective group of patients. METHODS: Dipyridamole-atropine (Dipatro: 0.84 mg/kg over 10 min i.v. dipyridamole with addition of up to 1 mg of atropine), dobutamine-atropine (Dobatro: up to 40 mcg/kg/min i.v. dobutamine with addition of up to 1 mg of atropine) and exercise (Ex, Bruce) were performed in 166 pts. Of them, 117 pts without resting wall motion abnormalities were enrolled in study (91 male; mean age 54 ± 10 years; previous non-transmural myocardial infarction in 32 pts, angina pectoris in 69 pts and atypical chest pain in 16 pts). Tests were performed in random sequence, in 3 different days, within 5 day period under identical therapy. All patients underwent coronary angiography. RESULTS: Significant coronary artery disease (CAD; ≥50% diameter stenosis) was present in 69 pts (57 pts 1-vessel CAD, 12 multivessel CAD) and absent in 48 pts. Sensitivity (Sn) was 96%, 93% and 90%, whereas specificity (Sp) was 92%, 92% and 87% for Dobatro, Dipatro and Ex, respectively (p = ns). Concomitant beta blocker therapy did not influence peak rate-pressure product and Sn of Dobatro and Dipatro (p = ns). CONCLUSION: When state-of-the-art protocols are used, dipyridamole and dobutamine stress echocardiography have comparable and high diagnostic accuracy, similar to maximal post-exercise treadmill stress echocardiography
